D-Aspartic Acid, as one kind chiral compound, plays great role in medicine, food etc, especially its important application prospect in pharmaceutical intermediates as the precursor of chiral drug synthesis. Currently, resolution of enantiomer is the main production method, of course, the enzymatic resolution is the most competitive. Application in pharmaceutical area In pharmaceutical synthesis, D-Aspartic Acid is mainly used as precursors and intermediates, such as the intermediate of Aspoxicillin (the first amino acid type penicillin in clinical use). At the same time, D-Aspartic Acid can be used in synthetizing D - Aspartate - Beta – Hydroxylamine(DAH), DAH DAH is a drug of virus infection, especially has resistant for some retroviruses and can be applied to treat AIDS, tumours etc. D-Aspartic acid joins to synthetize some antibiotic side chain, as the side chain of Beta-Lactam, it can improve the ant-bacter activity of antibiotic.The goods also can be used in synthesis of N-Methyl-D-Aspartate(NMDA, an important amino acid derivants, is the excitatory neurotransmitter L - glutamic acid homologue for mammalian central nervous system). The main route for NMDA is as below: D-Aspartic acid—(esterification) —D- Aspartic acid methyl ester hydrochloride—(DMF solvent, methyl iodide) —N-Methyl-D-Aspartic Acid Dimethyl Ester—(Base) —NMDA Application in food area D-Aspartic acid's acyl derivative has the function of resisting botulinus toxin, like myristica fragrans, D-Aspartic acid can replace sodium nitrite as a food preservative. In recent years, people gradually find and develop the application of D-Aspartic acid to enhance body function and immunity. In European and American countries, D-Aspartic acid has huge market prospect in health products. Now, we mainly adopt enzymatic resolution to product D-Aspartic acid, the commonly enzymes includes L-Asp-α- decarboxylase, L-Asp-β- Decarboxylase and Aspartase. The most mature method is using L-Asp-α- Decarboxylase to split. The enzyme can efficiently removal beta carboxyl generation L–Alanine from L-Aspartate that exists in DL-Aspartic acid, at the same time, it will have no effect on D-Aspartic acid, Thus D - aspartic acid was isolated.
